Overview

The purported effects of Tribulus terrestris are usually attributed to its steroidal saponins, chiefly protodioscin, which some laboratory and animal work suggests may influence androgen signalling, nitric-oxide release and erectile tissue; these mechanisms are not firmly established in humans [2]. Despite marketing as a testosterone booster, controlled trials in men generally show no meaningful rise in serum testosterone or in strength and body composition, indicating that any anabolic mechanism is weak or absent at the doses used [1].
In women, a placebo-controlled study reported improved sexual desire alongside increased free and bioavailable testosterone, hinting at a hormonal route in that group, though the effect and its mechanism remain uncertain [3]. The plant's beta-carboline alkaloids, harman and norharman, act on the nervous system; in sheep they accumulate in neurons and produce a lasting motor disturbance, which accounts for the neurological signs of Tribulus poisoning in livestock [4]. The same class of constituents underlies the photosensitisation seen in grazing animals, in which breakdown of chlorophyll-derived pigment in a compromised liver leads to skin reactions on exposure to sunlight [4].
