Overview

The peptide was first isolated from soybean in 1987, and its biological activity was later characterized by Alfredo Galvez in the laboratory of Ben de Lumen at the University of California, Berkeley.
Beyond soybean, lunasin has been identified in barley, wheat, and rye, and in grain crops such as amaranth, which broadens the range of foods that supply it. Interest in the peptide grew because diets rich in soy have been linked in population studies with lower rates of certain cancers, and lunasin emerged as one candidate factor behind that association [4].
Most of the evidence for lunasin comes from cell culture and animal work rather than humans. Early studies showed that it blocked chemical-carcinogen-induced transformation of cultured cells, bound preferentially to deacetylated histone H4, suppressed histone acetylation, and reduced skin tumor formation in mice [1]. Later research reported that it slowed the growth of non-small cell lung cancer cells by interfering with phosphorylation of the retinoblastoma protein and the cell cycle [2]. Review articles now describe lunasin as an epigenetic modulator with possible roles in both cancer prevention and treatment [3][4], and it has also been examined for effects on cholesterol and inflammation.
Direct human evidence remains thin. A small clinical trial that tested lunasin in people with amyotrophic lateral sclerosis found no benefit and noted digestive side effects. Lunasin is sold as a soy-derived dietary supplement rather than an approved medicine, and questions remain about how much intact peptide survives digestion and reaches tissues.
Mechanism
Lunasin is thought to act mainly at the level of chromatin, the packaged form of DNA. Its RGD motif lets the enter cells, after which it localizes to chromatin and binds preferentially to deacetylated histone H4, blocking the acetylation of histones H3 and H4. By preventing this acetylation it appears to push cells that are beginning to transform toward programmed cell death while largely sparing normal cells [1]. Additional work links lunasin to cell-cycle arrest, including reduced phosphorylation of the retinoblastoma protein and higher levels of the inhibitor p27, in cancer cell models [2], and to effects on integrin signaling [4].
