Apoaequorin is the protein component of aequorin, a calcium-sensitive, light-emitting protein originally isolated from the jellyfish Aequorea victoria. Aequorin was first isolated in the early 1960s by Osamu Shimomura, whose work on the jellyfish's luminous proteins also led to the discovery of green fluorescent protein and was recognized with a share of the 2008 Nobel Prize in Chemistry. Because its light output tracks calcium concentration, aequorin, and by extension apoaequorin, became a valuable reporter for measuring calcium signaling inside living cells [1].
Apoaequorin is marketed by Quincy Bioscience as the active ingredient of Prevagen, a dietary supplement promoted for memory and mild age-related cognitive decline. Independent assessments have been skeptical of these claims. A review in a drug bulletin and an analysis in a pharmacy journal each concluded that the clinical evidence is limited and does not convincingly support a memory benefit [2][3].
The company's own clinical study, the Madison Memory Study, did not show an overall benefit over placebo on its primary measures, and critics have noted reliance on selected subgroup findings. In 2017 the United States Federal Trade Commission and the New York Attorney General charged the marketers with false and deceptive advertising [3]. A further scientific objection is that apoaequorin is a protein, which would be expected to be broken down into amino acids during digestion rather than reaching the brain intact.

Mechanism
In its scientific role, apoaequorin works through the chemistry of aequorin. When apoaequorin is combined with coelenterazine and oxygen it forms a stable complex; the protein's EF-hand loops then bind calcium ions, triggering oxidation of the bound coelenterazine to coelenteramide and the release of carbon dioxide and a photon of blue light [1]. Because the amount of light depends on how much calcium is present, this reaction lets researchers track rapid changes in intracellular calcium. The mechanism proposed for the memory supplement, that ingested apoaequorin acts as a calcium-buffering protein in brain cells, is not well supported: as a protein it is expected to be digested into its constituent amino acids, and controlled evidence for a cognitive effect is lacking [2][3].
