Triamcinolone is a prescription corticosteroid. Local effects depend on the form and include skin thinning, lightening and visible vessels with topical use, and raised eye pressure or cataract with eye use. Injected depots can suppress the adrenal glands or cause Cushingoid changes if large or repeated. With systemic exposure it can add to the ulcer risk of NSAIDs and blunt the response to live vaccines, so it is used at the lowest effective dose for the shortest needed time.

The interaction with the most documented harm is with strong CYP3A4 inhibitors. Triamcinolone acetonide is cleared by CYP3A4, and ritonavir or cobicistat blocks that route so completely that a single joint or periradicular injection can produce iatrogenic Cushing's syndrome with adrenal suppression weeks afterwards; at least fifteen such cases have been published. Itraconazole, clarithromycin and grapefruit juice act by the same mechanism with less force. Strong inducers such as rifampicin, phenytoin and carbamazepine push the other way and can leave the steroid underactive.
The rest are class effects of a glucocorticoid. Combined with NSAIDs it raises gastrointestinal ulceration and bleeding risk; with potassium-wasting diuretics or amphotericin B it deepens hypokalemia; it opposes insulin and oral antidiabetic drugs by raising blood glucose; and it blunts the immune response to vaccines, which makes live vaccines hazardous during systemic or high-dose treatment.
