DOC (4-chloro-2,5-dimethoxyamphetamine) is a potent, very long-acting psychedelic amphetamine of the DOx family, bearing a chlorine at the 4 position. It acts as a 5-HT2A serotonin receptor agonist, with the shared 2,5-dimethoxy motif underpinning its receptor activity, and produces a long, sharply visual psychedelic state with a strong stimulant edge; in rodent studies it has shown rewarding and reinforcing effects. Like other DOx compounds it is metabolized partly by CYP2D6 and carries pronounced vasoconstriction and a marathon duration. Fatal intoxications have been reported, and interpreting blood concentrations in such cases is complicated by its high potency and the very low concentrations involved.

Mechanism
DOC is a 2,5-dimethoxy-4-chloro amphetamine. Its core action is agonism at the receptor, the receptor that drives the classic psychedelic state, behaving as a high-affinity partial there and at 5-HT2C. The amphetamine backbone slows metabolism and adds a stimulant component, and the persistent 5-HT2A action underlies its vasoconstriction and very long course.

DOC is potent at low milligram amounts and can last most of a day, so overshooting a dose is easy; it causes marked vasoconstriction, and overdoses have brought on hyperthermia (dangerous overheating), agitation, and seizures. It has appeared on blotter mislabeled as LSD. It should not be combined with MAOIs, vasoconstrictors, or other strongly serotonergic drugs because of serotonin toxicity and additive vessel narrowing.
