DOB (4-bromo-2,5-dimethoxyamphetamine) is a potent, very long-acting psychedelic amphetamine of the DOx family, bearing a bromine at the 4 position. It acts as an agonist at the 5-HT2A serotonin receptor, and the 2,5-dimethoxy substitution pattern is a key determinant of high-affinity binding and agonist activity at this site; in animals it produces 5-HT2A-mediated behaviors and serves as a training drug in drug-discrimination studies, with repeated dosing inducing tolerance accompanied by adaptation of cortical 5-HT2A and glutamate receptor binding. It is metabolized in part by CYP2D6 and, owing to its high potency, is commonly distributed on blotter. Its marathon duration, strong stimulant and body load, and pronounced vasoconstriction make it one of the riskier classic psychedelics.

Mechanism
DOB is a 2,5-dimethoxy-4-bromo amphetamine. Its core action is agonism at the receptor, the receptor that drives the classic psychedelic state, and it is a partial at 5-HT2A and 5-HT2C with unusually high affinity and slow receptor kinetics. The amphetamine backbone slows breakdown and adds a stimulant component; the strong, persistent 5-HT2A action on blood vessels drives its notable vasoconstriction.

DOB is extremely potent by weight and can last a full day or more, so misjudging a dose is easy and dangerous; its strong vasoconstriction has caused peripheral circulation problems, and overdoses can bring on agitation, hyperthermia (dangerous overheating), and seizures. Blotter mislabeled as LSD is a classic exposure route. It should not be combined with MAOIs, vasoconstrictors, or other strongly serotonergic drugs because of serotonin toxicity and additive vessel narrowing.
