Mechanism
Hyperforin has an unusual, multi-target pharmacology. In cell and tissue studies it inhibits the reuptake of a broad range of neurotransmitters, including , noradrenaline, , and , which is atypical for an antidepressant since most drugs target only one or two transporters [1]. Rather than blocking each transporter directly, hyperforin appears to work indirectly by activating TRPC6, a calcium-permeable cation channel in the cell membrane; opening this channel raises intracellular sodium and calcium, and the resulting rise in sodium reduces the driving force that transporters need to pull neurotransmitters back into the cell [1][2].
Hyperforin is essentially the only known selective opener of TRPC6, which has made it a research tool as well as a drug candidate. Through the same TRPC6 and calcium signaling it can promote neurite outgrowth and increase brain-derived neurotrophic factor signaling, effects linked to antidepressant action, and it also shifts the cellular balance of sodium, calcium, zinc and protons and influences function [1]. Separately, hyperforin binds and activates the pregnane X receptor, inducing cytochrome P450 enzymes such as CYP3A4, the basis of its many drug interactions [2].
