⚠️ HOW THIS DIFFERS FROM THE OTHER TWO MITOCHONDRIAL STACKS, stated first because they share roster. This one is about ENERGY PRODUCTION: getting fuel into the cycle and getting more ATP out of it. Mitochondrial management is about UPKEEP AND CLEARANCE, where spermidine's autophagy is the lead. Anti-neurodegeneration is about DAMAGE ACCUMULATION rather than production. PQQ and J-147 appear in more than one because mitochondria are where those stories genuinely meet, not because the rosters were padded.
J-147 enhances ATP synthase directly, and also raises BDNF and NGF with reported anti-anxiety and antidepressant-like effects. It appears in the anti-anhedonia stack for that same energetic reason: reward processing is expensive, and a metabolically tired brain feels flat even when nothing else is wrong.
PQQ is the mitochondrial biogenesis route via PGC-1alpha, which is a genuinely different lever from the other three and is worth naming as such. Building more mitochondria and running the existing ones better are not the same intervention.
TTFD, or fursultiamine, is a lipid-soluble thiamine derivative that crosses membranes far better than thiamine itself, which matters because thiamine's transporters saturate and ordinary supplementation hits a ceiling that TTFD does not.
⚠️ RESVERATROL IS THE ITEM TO BE HONEST ABOUT, and it is marked optional for that reason. Its human literature is far weaker than its popular reputation, its oral bioavailability is poor enough to make the cell-culture concentrations largely irrelevant, and several high-profile findings in the sirtuin field have not replicated. It is included because it belongs to the mechanism being described; it is not included because the evidence is good.
Every item here acts on mitochondria, but by four genuinely different routes, which is what makes this a stack rather than four overlapping bets on the same thing.
TTFD is the substrate-entry lever. Thiamine is the cofactor for pyruvate dehydrogenase and alpha-ketoglutarate dehydrogenase, which are the two gateways into and through the Krebs cycle; TTFD is a lipid-soluble derivative that crosses membranes far better than thiamine itself, which is the entire reason it exists. Nothing else here affects what gets in.
PQQ is the biogenesis lever, reported to drive PGC-1alpha signalling, which means more mitochondria rather than better operation of the existing ones. J-147 acts at the far end of the same pathway, on ATP synthase, the terminal enzyme of oxidative phosphorylation. Resveratrol is a signalling nudge through sirtuins and AMPK, which overlaps conceptually with PQQ's biogenesis story and is the reason it is the marked-optional item rather than a fourth independent lever.
Substrate in, more machinery, better throughput at the end, and a signal telling the cell to do more of all of it.
TTFD is the one item here with an acute character, and it is the reason people notice this stack at all: thiamine repletion in someone functionally short of it can be striking within days, and in someone replete it does approximately nothing. Which of those applies is not knowable in advance.
PQQ and J-147 are slow and mostly subclinical. Resveratrol should be expected to produce nothing perceptible, and its human literature is consistent with that.
Over months, the honest measure is fatigue tolerance rather than energy: how a long day ends rather than how a morning starts.
TTFD can produce a strong initial response in someone with a functional thiamine shortfall, including transient fatigue or irritability as metabolism shifts; that is well described and usually settles. One of TTFD's key papers is manufacturer-authored, and that flag is live on its entry. Resveratrol has meaningful CYP interactions and mild antiplatelet activity.
