⚠️ CURCUMIN'S REPUTATION IS FAR AHEAD OF ITS HUMAN EVIDENCE, and that gap is the honest headline of this stack rather than a footnote. It is one of the most-studied natural products in existence and most of that work is in cells, at concentrations an oral dose does not reach. It is on this list because the pairing with bromelain is exactly the response to that problem, not because the problem is solved.
Curcumin and bromelain are the pairing that makes this a stack rather than a list, and the reason is absorption rather than pharmacology. Curcumin's problem has never been potency; it is that oral bioavailability is close to nothing, which is why the literature is full of impressive cell work and thin human results. Bromelain is a proteolytic enzyme complex taken alongside partly for its own anti-inflammatory effect and partly because it is one of the classic co-administrations for getting more curcumin into circulation.
Resveratrol is deliberately NOT a second curcumin. It reaches inflammation through sirtuin activation and its own NF-kB effects, so stacking the two is additive rather than redundant, and it shares curcumin's absorption problem, which is why both are taken with fat.
SAM-e is the odd one out and is here on evidence rather than on mechanism-matching. It is a methyl donor rather than a plant polyphenol, and it carries the best human joint data of anything on this list.
Nothing here is a same-day effect, and a stack of anti-inflammatories that announced itself would be a worrying stack. Joint comfort, where it comes, comes over weeks.
SAM-e is the exception in both directions: it is the item most likely to be noticed, and what tends to get noticed is mood rather than joints, which is a real effect and also a reason to be careful with it.

SAM-e carries the other named risk: it can precipitate mania in bipolar disorder and it should not be combined with an SSRI, an SNRI, an MAOI or any other serotonergic drug without a doctor, because of serotonin syndrome. That is a hard stop rather than a caution. Bromelain is a pineapple derivative and a real allergen; latex-fruit cross-reactivity is worth knowing about before the first dose.
