All four are Khavinson short peptides from the St Petersburg Institute of Bioregulation and Gerontology. The claims attached to them are large: organ-specific gene regulation, telomerase activation, lifespan extension. The accessible evidence is mostly decades of Russian-language work that is hard to obtain, hard to appraise, rarely replicated outside the originating group, and largely unindexed in the databases this site verifies against.

That is not a reason to refuse the stack. It is the reason the honest framing is a coherent research programme with a devoted following and a thin external evidence base, rather than a list of mechanisms stated flatly.

Epitalon is a tetrapeptide, Ala-Glu-Asp-Gly, and the best known of the four. Its telomerase claims are the most widely repeated and the least independently verified; it also appears in the nasal mood stack on the pineal and circadian angle rather than on any antidepressant data.

Pinealon is a tripeptide, Glu-Asp-Arg, proposed as a brain and pineal bioregulator with claims about neuronal protection and cognitive function.

Cortagen is a tetrapeptide, and ⚠️ the sources disagree about what it is for: some describe peripheral nerve, others the adrenal cortex axis. That disagreement is itself informative about how loosely specified this literature is.

Vesugen is a tripeptide, Lys-Glu-Asp, proposed as a vascular bioregulator.

On practicalities: these are short peptides with poor oral bioavailability, which is why they are typically injected subcutaneously or taken sublingually. Several are candidates for intranasal preparation, and the route is worth stating rather than leaving implied.

how they work together

The proposed synergy is organ specificity: each peptide is claimed to act on a different tissue, so running four is claimed to address four systems rather than one system four times. Pinealon toward brain and pineal, cortagen toward nerve or adrenal cortex, vesugen toward vasculature, epitalon toward the pineal and telomerase axis.

⚠️ THAT IS THE CLAIM RATHER THAN A DEMONSTRATED MECHANISM, and the distinction matters more here than anywhere else on this page. The proposed mechanism is that these short peptides enter the nucleus and bind DNA directly, modulating gene expression in a tissue-selective way. That is a large claim for a tripeptide, the supporting work comes almost entirely from the originating institute, and it has not been reproduced independently in a form this site can verify.

What can be said honestly is that these four are used together because the programme that produced them treats them as a set, and that the internal logic of that programme is consistent. Whether the logic corresponds to anything is a separate question and is not settled.

Reports are quiet and are dominated by sleep quality, which is unsurprising given the pineal framing and is also the effect most susceptible to expectation.

These are typically run in short courses of ten to twenty days rather than continuously, which is the programme's own convention, and reported effects are described as building across a course.

Anyone expecting the lifespan and telomere claims to manifest as a sensation will find nothing, because those claims are about processes with no perceptual correlate at any timescale.
