⚠️ THIS IS THE THIRD STACK IN A FAMILY AND THE DISTINCTION HAS TO BE REAL, so here it is: mitochondrial longevity is about production, anti-neurodegeneration is about damage accumulation, and this one is about upkeep and clearance. Spermidine is the item that makes that true, which is why it leads rather than sits fourth.
Spermidine is a polyamine, present in food and produced endogenously, that induces autophagy; wheat germ is the usual source in supplement form. Its longevity literature is largely observational and animal work, and the human trials that exist are small. That is worth saying, because spermidine currently occupies the position resveratrol occupied a decade ago.
PQQ drives mitochondrial biogenesis via PGC-1alpha. J-147 acts on ATP synthase and raises BDNF and NGF.
⚠️ NADH IS NOT NAD+, AND THIS IS THE MOST CONFUSED CORNER OF THE LONGEVITY AISLE. They are the reduced and oxidised halves of the same couple and they are not interchangeable in a supplement context. Most products sold in this space are neither: they are precursors, usually nicotinamide mononucleotide or nicotinamide riboside, which the body converts onward. NADH itself is poorly absorbed orally and is degraded by stomach acid, which is why stabilised or sublingual forms exist and why the absorption question is unsettled rather than solved. The anti-neurodegeneration stack asks for NAD+ specifically and that is a different item, deliberately.
Build, run, fuel and clear. The first three are covered by every mitochondrial stack; the fourth is what this one is actually for.
Spermidine is the lead. It is a polyamine that induces autophagy, which in the mitochondrial context means mitophagy: the selective removal of mitochondria that have become damaged. That is a categorically different intervention from the other three. Building more mitochondria and running them harder both increase capacity; clearing broken ones improves the average quality of what is there. A population of mitochondria that is never culled degrades regardless of how many are added.
PQQ supplies the build side through PGC-1alpha, which pairs with spermidine in a specific way: clearance without replacement reduces total capacity, and replacement without clearance accumulates damage. Running both is the coherent version.
J-147 improves throughput at ATP synthase, and NADH is the reduced electron carrier that the respiratory chain consumes at complex I. Those two sit at opposite ends of the same chain.
Spermidine and PQQ produce nothing perceptible, and the honest framing is that autophagy is not a sensation. Anyone expecting to feel mitophagy will conclude the stack does not work.
NADH is the item most likely to produce something acute, and reports vary widely, which is consistent with its absorption being poor and variable.
This is a months-and-years stack presented as a daily one, and the useful expectation is no expectation. That is unsatisfying and it is accurate.
Spermidine's human evidence is thin relative to its reputation and rests largely on observational and animal work. NADH's oral absorption is poor and contested; a product's form matters more here than in almost any other supplement.
