Butyric acid (butanoic acid) is a short-chain fatty acid with the formula CH3CH2CH2COOH, present in the body mainly as its salt, butyrate. It is produced when gut bacteria ferment dietary fiber in the colon, and it also occurs naturally in butter and other dairy fats. Butyrate serves as the primary energy source for the cells lining the colon and is widely studied for its roles in gut and metabolic health.
L-Threonine is one of the nine essential amino acids; the body can't make it, so it has to come from diet. It's a building block for protein and a source of glycine and serine, and the gut uses a large share of dietary threonine to build mucin, the protective mucus layer that lines the intestine. It's sold as a supplement, though for most people a normal protein intake already covers the requirement.
Larazotide (larazotide acetate, formerly AT-1001) is a first-in-class orally administered eight-amino-acid peptide that regulates intestinal tight junctions to restore barrier function. In celiac disease this limits translocation of immunogenic gliadin fragments across the epithelium, and the peptide acts locally with minimal systemic absorption; a large randomized trial found the lowest 0.5 mg dose reduced symptoms in patients who remained symptomatic despite a gluten-free diet, and it advanced into phase 3 development. Beyond celiac disease, larazotide has restored barrier integrity in models of intestinal ischemia through repair of claudin-4-sealed junctions and has been explored for other conditions linked to increased gut permeability.
Tributyrin, also called glyceryl tributyrate, is a triglyceride made of glycerol esterified with three molecules of butyric acid, and it occurs naturally in butter. It serves as a stable, orally absorbed prodrug that releases butyrate into the body, letting the short-chain fatty acid reach the bloodstream more effectively than butyrate salts [1]. Because butyrate acts as a histone deacetylase inhibitor, tributyrin has been studied in cancer, gut health and, more recently, as a postbiotic dietary supplement [4].
