After an oral dose, gas chromatography and mass spectrometry recover boldenone itself plus nine metabolites from urine, most of them 5-beta reduced; more than 96% leave as stable conjugates rather than as the readily hydrolysed glucuronides that routine screening looks for [5]. An earlier study of the same 1-dehydro series in humans reached compatible conclusions [6].
- One of those human metabolites is androsta-1,4-diene-3,17-dione, the 1-ene counterpart of androstenedione [5]. Androstenedione is the ordinary substrate that aromatase converts to oestrone [17], and the enzyme's active site is shaped around that androgen skeleton [18]; what the extra 1,2 double bond does to the product is the open question, and is not settled by the metabolism work itself.
- A reassuring oestrogen result on boldenone is weaker evidence than it looks. Direct estradiol and testosterone immunoassay kits cross-react with other steroids [20][21] and overstate low concentrations, which is why mass spectrometry is the reference method [22]. On a compound whose metabolites are themselves unusual steroids, an immunoassay number is not a measurement of what a reader thinks it is.
- Raised creatinine on boldenone does not by itself mean kidney injury; serum creatinine tracks muscle mass and physical activity independently of kidney function [27].
