Hydrocortisone acetate is the acetate ester of hydrocortisone (cortisol), a glucocorticoid corticosteroid used mainly as an anti-inflammatory and anti-allergic agent. It is applied to the skin for irritation, itching and eczema, given rectally for hemorrhoid symptoms, and injected into joints or soft tissue to calm inflammation. Adding an acetate group at the steroid's 21-position slows its breakdown and changes how readily it is absorbed.
Tacrolimus, originally known as FK-506, is a macrolide immunosuppressant of the calcineurin-inhibitor class. It was discovered in the 1980s from a soil bacterium and is used mainly to prevent rejection after organ transplantation and, as an ointment, to treat eczema. By blocking a signaling pathway in T cells, it suppresses the immune responses that would otherwise attack transplanted tissue.
